Urine Culture

 Intro:

This test required to diagnose a Urinary Tract Infection such as pain and burning when urinating and frequent urge to urinate. Antibiotic therapy may be prescribed without requiring a urine culture for symptomatic young women who have an uncomplicated lower urinary tract infection. If there is suspicion of a complicated infection or symptoms do not respond to initial therapy, then a culture of the urine is recommended. Pregnant women without any symptoms may be screened for bacteria in their urine, which could affect the health and development of the fetus.

Lab test for Urine Culture:

Important notes before testing:

-Patient must not take any antibiotics before testing by 48 hours.
-First morning void is ideal specimen.
-The sample must be in the lab for testing within 30 minutes after taking.

Steps for testing:
1.Mix the sample well before taking your specimen for culturing.
2.Swap your specimen on the Blood agar plate first then by sterile wire loop swap on the MacConkey agar (Mac) or EMB agar.
3.Leave it for 24 hours at 37ºC.

Results:
-If cultures grown on the both plates so the bacteria is G +ve and if grown on Mac agar only so it is G-ve bacteria.
-Appearance of different types of bacteria on the plate indicates Contamination during culturing so you must repeat the culturing again.
-The presence of white blood cells and low numbers of microorganisms in a symptomatic patient is a condition known as acute urethral syndrome.
-Notice the shape on colonies and the arrangement.
-Make your Gram stain slide to test under microscope.

Types of bacteria expected:
Staphylococcus - Streptococcus - Neisseria - N.meningitidis - N. gonorrheae - Enterobacter- E . Coli - Klebsilla - Citrobacter - Salmonella - Shigella - Proteus - Pseudomonas.

Normal:
Organisms < 1,000 organisms per ml

Abnormal:
Organisms >10,000 per ml suggests UTI.

Organisms >1,000 per ml needs repeat urine culture.
