Toluidine blue staining procedure

Reagent

150 mg of Toluidine blue dissolve in PBS buffer.
Hank's solution (Difco)
PBS buffer: phosphate buffered saline solution (Sigma)

Technique

1. In a test tube, add 3 drops of sediment.
2. Add 1 ml of PBS buffer or Hank's solution (Difco)
3. add 1 or 2 drops of stain depending on cellularity
4. Centrifuge
5. Aspirate the added fluid volume and read
